Scientists Pinpoint Gum Disease Pathogen As Possible Cause Of Alzheimer's

These findings, published online January 24 in the journal Science, indicate that lack of sleep alone helps drive the disease, and suggests that good sleep habits may help preserve brain health.

New research identified P. gingivalis, the same bacteria responsible for gum disease, as the possible culprit behind Alzheimer's disease.

"Not enough people are asking what is upstream of the plaques ... and [brain] inflammation", said Lynch, who has a background in Alzheimer's research and was frustrated by the string of failed therapies for the disease.

Last year, a Taiwanese study found that people with a 10-year or longer history of chronic periodontitis (CP) were 70% more likely than people without the condition to develop Alzheimer's. "We will have to see the outcome of this ongoing trial before we know more about its potential as a treatment for Alzheimer's", he said.

The researchers found that in mice, which are nocturnal creatures, tau levels in the fluid surrounding brain cells were about twice as high at night, when the animals were more awake and active, than during the day, when the mice dozed more frequently.

Lynch mentioned that despite "significant funding and the best efforts of academic, industry and advocacy communities, clinical progress against Alzheimer's has been frustratingly slow". "He points out that while most of the data presented in the Cortexyme study supported their hypothesis, gingipains weren't found in all of the Alzheimer's-affected brains, 'so whilst it may be a cause, the data don't exactly support it being the only cause". One widely believed theory is that the disease is driven by the production and deposition of beta-amyloid plaques between neurons in the brain. He cited previous research that proved other factors are also responsible for the development of the disease.

Alzheimer's is a much-feared condition that, according to the Alzheimer's Association affected over 5.7 million people in the United States alone, in 2018. A vaccine for gum disease would be welcome - but if it also stops Alzheimer's the impact could be enormous.

The group is believed to be the largest one yet to cross the border in a bid to get asylum in the U.S. President Trump and congressional Democrats are now deadlocked over funding for a border wall.

One theory promoted by the new research is that cognitive impairment is causing adverse oral health.

While the patient groups had similar overall AD pathology, the authors found, Hispanics with AD showed greater small blood vessel disease in the brain than non-Hispanics with AD, as well as increased amyloid angiopathy, the accumulation of protein fragments in blood vessels associated with AD. For the study, chemists developed a drug to help control these modifications, and injected lab mice with the compound three times over three days.

People with Lewy body dementia and Huntington's disease tend to have high levels of the protein in the blood. "But the evidence of causation hasn't been convincing".

"Now, for the first time, we have solid evidence connecting the intracellular, Gram-negative pathogen, P. gingivalis, and Alzheimer's pathogenesis".

In addition to Pg itself, the study team also detected the organism's toxic proteases, or gingipains, in the neurons of patients with AD. This timeline gives researchers an opportunity to study what happens in the brain in the years before cognitive symptoms arise. Should wakefulness increase, however, tau can accumulate faster than it can be cleared away, and then tau-a hallmark of Alzheimer's disease-may stay elevated, form toxic clumps, and spread throughout the brain, accelerating neurodegeneration.

Testing how NfL levels change could also become a useful way to develop new Alzheimer's treatments.
